Since that time, this technique has been trustworthy tool to review necessary protein localization and cytoskeletal company in cortex of unfertilized eggs and embryos during first cleavages. This study was aimed to estimate the reliability of RT-qPCR to evaluate levels of maternal transcripts which can be localized in egg cortex. Firstly, we selected seven possible research genes, 28S, Cycb, Ebr1, GAPDH, Hmg1, Smtnl1 and Ubb, the transcripts of which are maternally deposited in ocean urchin eggs. The prospect reference genes were ranked by five various algorithms (BestKeeper, CV, ΔCt, geNorm and NormFinder) predicated on calculated level of stability both in eggs aswell as separated cortices. Our results revealed that gene standing differs in total RNA and mRNA samples, though Ubb is the most suitable reference gene both in cases.
